Lion’s Mane, or Hericium erinaceus, is one of the best-known functional mushrooms in the supplement market. Products based on it are frequently promoted to people seeking support for memory, concentration, mental clarity and long-term cognitive function.
There is a scientific basis for this interest. Lion’s Mane contains bioactive compounds investigated for their influence on neuronal growth, survival and communication. Several small human trials have also reported encouraging results in cognitive performance, stress and mood.
However, “Lion’s Mane” on a label reveals little about what is actually inside a supplement. A mushroom powder, hot-water extract, mycelium product and extract concentrated for specific compounds can all have different chemical profiles.
This article examines what science says about Lion’s Mane and cognitive function, and how consumers can identify products whose quality makes the proposed benefits scientifically plausible.
Hericenones, NGF and brain support
Two groups of compounds appear frequently in Lion’s Mane research: hericenones and erinacines. Hericenones are found mainly in the fruiting body, while erinacines are associated primarily with the mycelium.
Hericenones have attracted attention because of their potential relationship with nerve growth factor, or NGF. NGF supports the growth, maintenance and survival of nerve cells. Laboratory research has found that compounds and extracts derived from Lion’s Mane can influence NGF-related activity, offering a possible explanation for the mushroom’s association with cognitive support.
Another relevant protein is brain-derived neurotrophic factor, or BDNF, which is involved in learning, memory and neuronal survival. Research connecting Lion’s Mane with BDNF is promising, although much of it concerns erinacines or mixed extracts rather than hericenones alone.
Some Lion’s Mane compounds may also have properties that allow them to cross the blood–brain barrier. Evidence is stronger for certain erinacines than for hericenones, so this should be described as plausible and under investigation rather than proven for every product.
Why not every Lion’s Mane product is equivalent
Two supplements can both display “Hericium erinaceus” on the label while containing very different amounts of the compounds investigated in cognitive research.
A powdered fruiting body may contain hericenones, but their concentration can be low, variable or unknown. A hot-water extract may mainly concentrate polysaccharides such as beta-glucans. These are valuable compounds, often studied in relation to immunity, but a high beta-glucan result does not prove meaningful hericenone content.
Other methods, including supercritical CO₂ extraction, can recover more lipophilic compounds and create products designed around hericenones. No method is automatically best for every purpose. The key question is whether it suits the compounds the producer claims to provide.
Consumers interested in cognitive support should make two checks. First, look for a laboratory report showing the concentration of relevant compounds, ideally for a specific batch. A percentage or milligrams-per-gram result is more useful than a statement that a product simply “contains hericenones”.
Second, check the analytical method. High-performance liquid chromatography, or HPLC, is used to separate, identify and quantify individual compounds in complex extracts. A credible report should state the method, measured compounds, units, test date and, ideally, the laboratory and batch number.
“Many powdered Lion’s Mane products mention hericenones but do not state how much is actually present. A quality extract should provide a meaningful, quantified amount – potentially several per cent – and confirm it using a trustworthy analytical method such as HPLC,” says Piotr Pałucki, founder of ExtractHome, a manufacturer of Lion’s Mane and ashwagandha extracts.
What do human studies show?
Human research remains limited, but several studies have produced encouraging findings.
Mild cognitive impairment
A double-blind, placebo-controlled trial involved 30 Japanese adults aged 50 to 80 with mild cognitive impairment. Participants received Lion’s Mane or placebo for 16 weeks. The Lion’s Mane group achieved significantly better cognitive scores at weeks eight, 12 and 16. Scores declined after supplementation ended, suggesting that continued intake may have contributed to the result.

Healthy older adults
A 2019 randomised, double-blind, placebo-controlled study examined 12 weeks of Lion’s Mane supplementation in healthy adults over 50. Researchers observed a significant benefit in the Mini-Mental State Examination, although other cognitive tests did not show equally clear differences. The findings were positive but selective.

Cognitive performance and stress
A 2023 pilot study investigated Lion’s Mane in 41 healthy adults aged 18 to 45. After a single dose, the supplement group completed the Stroop test more quickly. After 28 days, researchers also observed a tendency towards lower subjective stress. Not every outcome improved, but the findings suggest potential relevance for younger adults as well as older populations.

Mood and psychological wellbeing
In another placebo-controlled study, 30 women consumed Lion’s Mane or placebo products for four weeks. The Lion’s Mane group reported improvements in measures related to depression, anxiety and psychological wellbeing. Although the study did not directly test memory or concentration, it adds to evidence of broader neurological effects.

What consumers should take from the evidence
The evidence does not show that every Lion’s Mane supplement will produce the same results, but it offers credible reasons for interest.
Lion’s Mane contains compounds associated with neurotrophic pathways, while early human trials have reported improvements in selected measures of cognition, stress and mood. The research is developing, but the signal is meaningful.
The practical conclusion is that the mushroom’s name alone is not a quality standard. A product intended to support cognitive function should provide transparent information about the mushroom material used, extraction method, relevant compounds and their concentration.
Supplements backed by batch-specific documentation and reliable methods such as HPLC provide a stronger basis for informed use than products relying only on optimistic claims. The key question is not simply whether a supplement contains Lion’s Mane, but whether it contains a measurable, well-documented profile of the compounds that made the mushroom scientifically interesting.
